Abstract

The invention comprises secondary and tertiary amines of the general formula: <FORM:0848690/IV(b)/1> (wherein Q represents a saturated or unsaturated aliphatic or mixed aliphatic-alicyclic radical composed of two or three isoprene residues and bound by a single or double bond to R3, which represents <FORM:0848690/IV(b)/2> R1 represents hydrogen or an alkyl or aralkyl radical, and R2 represents hydrogen (except when R1 is hydrogen), or an alkyl, aryl or aralkyl radical in which one hydrogen atom may be replaced by a free or etherified hydroxy group, or an alicyclic, mixed aliphatic-alicyclic, heterocyclic or mixed aliphatic-heterocyclic radical, or R1 and R2 form a heterocyclic ring with the nitrogen atom), and therapeutic, non-toxic acid addition salts thereof, and the preparation thereof by: (a) reacting a ketone of the general formula: <FORM:0848690/IV(b)/3> with an amine of the general formula HNR1R2, or a primary or secondary amine of the general formula Q-R3-NH-R2 with an aldehyde or ketone having the same carbon skeleton as the radical R1, either in the presence of formic acid, with subsequent hydrolysis of the formyl compound first produced, or in the presence of hydrogen and a metal hydrogenation catalyst (in which case simultaneous saturation of double bonds may occur when non-saturated starting materials are employed); (b) reacting a halide of the general formula Q-R3-Hal with a primary or secondary amine of the general formula HNR1R2, or a primary or secondary amine of the general formula Q-R3-NHR, with a halide of the formula R2-Hal; or (c) acylating a primary or secondary amine of the general formula Q-R3NH-R2 with an organic and, acid halide or acid anhydride so chosen as to produce an acylated amine of the general formula: <FORM:0848690/IV(b)/4> (wherein R11 represents an acyl group having the same carbon skeleton as the radical R1) and reducing this product by means of a complex metal hydride. Any double bonds still present in the radical Q-R3- may be subsequently reduced wholly or partly by means of hydrogen and a noble metal hydrogenation catalyst, e.g. in an acidic medium. Reference is also made to the preparation of N-[3-(p-hydroxyphenyl)-sec.-butyl] -tetrahydroionylamine hydrobromide by boiling the corresponding methoxy base with hydrobromic acid. The products are useful as spasmolytics. Tetrahydroionyl bromide, is prepared by the action of dry hydrogen bromide on tetrahydroionol, itself obtained by hydrogenation of b -ionone to tetrahydroionone and reduction of this with LiA1H4.
