Abstract
Tetracycline free base in a pure form having improved colour characteristics is recovered from solutions thereof by contacting a methyl alkyl ketone solution of the crude tetracycline with acidulated water at a pH of 1.5 to 2, separating the resulting aqueous phase containing the tetracycline from the solvent phase, adjusting the pH of the aqueous phase to pH 2 to 4 and preferably 3.5, and recovering the tetracycline by crystallization from the aqueous phase. It is preferred that the quantity of acidulated water be small and that the pH of extraction be 1.8 to 2. Specified methyl alkyl ketones are methyl isobutyl ketone p (preferred), methyl butyl ketone, methyl propyl ketone, methyl isopropyl ketone and methyl amyl ketone. A preferred methyl isobutyl ketone solution of tetracycline is that obtained by extraction from a fermentation broth in the manner described and claimed in Specification 821,310. A methyl alkyl ketone solution may be prepared by extracting at pH 1.5 the filter cake obtained by adjusting a fermentation broth to pH 7 to 8.5 and filtering. The tetracycline free base may be used therapeutically without further purification; or may be treated with butanol, 2-ethoxyethanol and hydrochloric acid to form the hydrochloride. Reference has been directed by the Comptroller to Specification 747,779.
