Abstract
Carboxamides of the general formulae <FORM:0826668/IV (b)/1> <FORM:0826668/IV (b)/2> and their acid addition salts (R3 and R4 represent hydrogen atoms or alkyl groups, R5 represents an alkyl group containing from 1 to 5 carbon atoms, and of the symbols X, Y and Z two are hydrogen or alkyl (and may be different) and the other represents the group -CONR1R2 wherein R1 is aryl which may be substituted e.g. by alkyl or alkoxy groups of from 1 to 7 carbon atoms, and R2 is hydrogen) are prepared by forming by mixed anhydride synthesis a carboxamide of formulae <FORM:0826668/IV (b)/3> <FORM:0826668/IV (b)/4> reacting the carboxamide with an alkylating agent R5A (A is an acid residue) and then selectively hydrogenating the product. Detailed examples illustrate the preparation of N - methylhexahydropicolyl - 2,6 - dimethylanilide hydriodide, N - methylhexahydroisonicotinyl-2,6-dimethylanilide hydrochloride, Nmethylhexahydronicotinyl 2,6-dimethylanilide hydriodide, N - ethylhexahydroisonicotinyl2,6 - dimethylanilide hydrochloride and Nmethyltetrahydroquinaldinyl - 2,6 - dimethylanilide and its hydrochloride. Specification 770,129 is referred to.ALSO:Anaesthetic compositions comprise carboxamides of the formulae <FORM:0826668/VI/1> and their acid addition salts (R3 and R4 are hydrogen or alkyl groups, R5 is alkyl containing less than 6 carbon atoms, two or X, Y and Z are hydrogen or alkyl and the third is -CONR1R2, where R1 is aryl and R2 is hydrogen) together with a pharmaceutical carrier, e.g. an aqueous or non-aqueous solvent for injection, or an ointment base for topical application.
