Abstract

The invention relates to a crystalline calcium glucoheptonate complex of streptomycin or dihydrostreptomycin of formula <FORM:0822081/IV (a)/1> wherein (Str) is streptomycin or dihydrostreptomycin. The complex is produced by reacting streptomycin sulphate or dihydrostreptomycin sulphate with an equivalent amount of calcium glucoheptonate in water, concentrating below 40 DEG C. and adding a water-miscible organic solvent to precipitate the complex. 6 to 8 litres of water are employed for each mol. of complex and the resulting solution is diluted with 4 to 6 volumes of the organic solvent which may be an aliphatic alcohol, especially methanol, dioxane, acetone or a mixture thereof. The calcium glucoheptonate may be produced in situ by adding glucoheptonic acid to an aqueous suspension of calcium hydroxide at pH 5 to 7. Glucoheptonic acid is a mixture of d-gluco-d-gulo-heptonic acid or d-glucoheptonic acid with minor amounts of d-gluco-d idoheptonic acid or beta-d-glucoheptonic acid. The complexes show a low chromic toxicity and are used in pharmaceutical preparations (see Group VI).ALSO:A crystalline calcium glucoheptonate complex of streptomycin or dihydrostreptomycin having the formula <FORM:0822081/VI/1> , wherein (Str.) is streptomycin or dihydrostreptomycin (see Group IV (b)), having a low chronic toxicity, is employed in a dosage of 1 to 5 grms. in pharmaceutical preparations for injection together with a diluent such as sterile water or saline solution. Saline solutions may also be used in the form of aerosols. Other streptomycin and/or dihydrostreptomycin salts may be present in proportions of 5 to 95% of the antibiotic content of the preparation. Specified are the sulphate, pantothenate, p-aminosalicylate or the salt with pyruvic acid isonicotinyl-hydrazone. Optional additions are: (a) water-soluble calcium salts such as calcium ethylene diamine-tetra-acetate; (b) antibiotics, e.g. penicillin; (c] other drugs; and (d) thickening, stabilising, wetting and buffering agents.
