Abstract

Nucleic acid is extracted from animal and vegetable materials, e.g. fresh fish milt, by treating the broken-up material with caustic alkali solution, precipitating the alkali metal nucleate by addition of an alcohol, mixing the precipitate with an aqueous suspension of calcium sulphate, heating and filtering the mixture and precipitating substantially pure nucleic acid by addition of a mixture of an alcohol and hydrochloric acid to the filtrate. The mixture of the precipitate and calcium sulphate may be first hot-filtered and then cold filtered. The nucleic acid may be suspended in alcohol and mixed with a solution of a metal salt in alcohol to form, by double decomposition, the nucleate of the metal employed. The alcohols used are lower aliphatic alcohols containing not more than 4 carbon atoms. Solutions of sodium, manganese, copper or iron sulphates, or calcium, magnesium or cobalt chlorides are specified.
