Abstract
a -Substituted isonicotinic thioamides, where the substituent is alkyl, aryl, or aralkyl, are prepared by the following series of reactions: (a) the ketone RCOCH2 is condensed with an oxalic acid diester in the presence of an alkali metal alcoholate to form an acyl-pyruvic acid ester; (b) the latter is condensed with cyanacetamide in the presence of a nitrogeneous base such as piperidine to form an ester of 6-R-3-cyano-4-carboxy-2-pyridone, which is (c) treated with hot concentrated HCl to form 6-R-4-carboxy-2-pyridone; (d) chlorination with POCl3+PCl5 gives the 6-R-2-chloro-4-acylchloride, which is then extracted with alcohol and esterified; (e) catalytic hydrogenation removes chlorine to form the a -substituted isonicotinic acid ester; (f) the ester is treated with NH3 to form the acid amide, which is treated with P2O5 to give the nitrile; (g) the nitrile, in an anhydrous solvent such as ethanol, is treated with H2S, in the presence of a tertiary nitrogenous base, such as triethanolamine. In an alternative procedure for the case of a -methyl-isonicotinic thioamide, pyruvic acid is condensed with NH3 in an alcoholic medium to form uvitonic acid, which is decarboxylated at about 300 DEG C. to a -methyl-isonicotinic acid; the acid is esterified, and the ester then treated by steps (f) and (g) above. In examples, the a -substituent is methyl, ethyl, n-propyl, isopropyl, n-butyl, isobutyl, amyl, n-hexyl, phenyl and benzyl.
