Abstract
L(+)-Glutamic acid is prepared by cultivating a strain of a species of Cephalosporium in a nutrient medium containing urea and a source of carbohydrate under aerobic conditions until appreciable quantities of said glutamic acid are formed in the medium. The Cephalosporium strain is preferably one of the following species: C. salmosynnematum, C. diospyri and C. acremonium. Specified carbohydrate sources are sucrose, glucose, corn syrup, molasses and potato, corn or wheat starch. The urea may be supplemented by nitrates, ammonium salts, protein hydrolysates and proteins such as wheat gluten, corn gluten meal, soybean meal, distillers' solubles and cornsteep liquor. Salts such as sodium chloride, magnesium sulphate and potassium sulphate may also be added to the nutrient medium. The urea concentration is preferably 0.1 to 5 per cent by weight of the reaction medium and the urea may be added at the beginning of or during the fermentation. The temperature is preferably maintained between 25 DEG and 32 DEG C. Aerobic conditions may be maintained by passing through 0.5 to 2.5 volumes of air per volume of medium per minute. The glutamic acid is isolated from the fermentation medium by chromatographic adsorption or by extraction with a polar solvent. Specification 776,722 is referred to.
