Abstract

Vitamins of the B12 group are purified by means of partition chromatography wherein the chromatographic column is filled with cellulose and the developer used comprises a single phase elution agent which consists of aqueous n-butanol containing not more than 18 per cent by volume of water or of aqueous secondary butanol containing not more than 31 per cent by volume of water. Enhanced results are obtained by effecting the development in the presence of neutral and readily dissociating salts, the anions of said salts being monobasic and consisting of at least four non-metal atoms, e.g. the sodium or potassium salts of the following acids: perchloric acid (HClO4), chloric acid (HClO3), bromic acid (HBrO3), nitric acid (HNO2), hydrogen tetraphenyl boron (H-B (C6H5)4 \sH ), monochloroacetic acid (Cl.CH2.-COOH), dichloroacetic acid (Cl2CH.COOH), trichloroacetic acid (Cl3C.COOH), monobromoacetic acid (BrCH2COOH), dibromoacetic acid (Br2CH.COOH), camphosulphonic acid, benzenesulphonic acid (C6H5SO3H), 2,4-dichlorobenzoic acid (C6H3Cl2COOH), sulphanilic acid (NH2.C6H4.SO3H) and a -naphthalene sulphonic acid. These salts may be admixed with the developer or with the chromatographic column itself and may be used in a concentration of 0.002 to 0.02 mols. per litre of developer. The cellulose column may be in powder or in flake form. The developer may contain a small quantity of hydrocyanic acid. Examples describe the separation of B12 vitamins from digested sludge water saturated n-butanol and sec.-butanol. Specification 748,043 is referred to.
