The invention comprises 6-chloro- and 6-iodopurine, and also their preparation by heating hypoxanthine with phosphoryl chloride at a temperature above the boiling point of the latter but below 200 DEG C. and, if 6-iodopurine is required, reacting the resulting 6-chloropurine with hydriodic acid. Suitably the 6-chloropurine is made by heating hypoxanthine with phosphoryl chloride in the presence of N : N-dimethylaniline for 2 to 5 hours or by heating under pressure with a reagent comprising phosphoryl chloride with 1 to 10 per cent of water at 140 DEG C. to 200 DEG C. for 2 to 8 hours.
