Abstract
The invention comprises 1:5 naphthyridine derivatives having alkoxy in 6-position, alkyl in 2-position and hydroxy in 4-position. They may be obtained by the reaction between a 6-alkoxy-3-amino-pyridin with an ester having, in one tautomeric form, the formula XO - CR1 = CR4.COOY in which X and R1 are H or an alkyl group, R4 is H or the group COOY and Y is an alkyl group, whereby a b -pyridylaminoacrylic ester is produced, and ring-closing by elimination of alcohol, hydrolysing the carboxylic ester group produced, when present, and decarboxylating if required. The 4-halogen derivative may be obtained by the action of known halogenating agents on the 4-hydroxy compound. In examples 6-n-alkoxy-3-aminopyridines are condensed with ethylacetoacetate- or ethoxymethylene malonic ester, to give the corresponding b -(6-alkoxy-pyridyl-3)-aminocrotonic ester or b - (6 - alkoxypyrid - 3 - ylamino) - a - carbethoxy acrylic ester, which products are cyclized by heating, hydrolysed if required, decarboxylated also if required and converted into the 4-chlor-1:5 naphthyridines having the 6-alkoxy group, to correspond, by means of POCl3. Intermediate products specified include 4-hydroxy - 6 - methoxy - 3 - carbethoxy 1:5-naphthyridine, and the corresponding 3-carboxylic acid.
