665,073. Coatings for acid-sensitive drugs. LOEWE RESEARCH FOUNDATION, Inc., J. Nov. 2, 1949 [Nov. 10, 1948; June 3, 1949], Nos. 28083/49 and 28084/49. Class 81 (i). An acid-sensitive drug is prepared in a form suitable for oral administration by incorporating said drug with an anion exchange resin, which is mixed with the drug or used as a coating therefor. Preferably the drug is first dispersed in a gel and the resin incorporated therein or coated thereon. Such a gel may be further coated with a carboxy alkyl cellulose ether. Specified drugs are penicillin salts, e.g. the sodium, aluminium, iron, magnesium and cobalt salts; penicillin, aureomycin, chloromycetin, streptomycin, bacitracin, subtilin, polymyxin, dihydrostreptomycin; hormones, e.g. estrogens; insulin, adrenalin and heparin. The anion-exchange resin may be natural or synthetic, e.g. aminoformaldehyde resins such as aniline formaldehyde, amino-mono-saccharide condensates, guanidine-formaldehyde resins, alkali treated asphalt and the resins disclosed in French Specification 820,969 and Specification 489,173, [Group IV]. Gel-forming colloids are natural gums; agar, starch, gelatine, casein, soya bean protein, yeast protein, blood protein and albumin. The preparation may be in the form of a pill, tablet or capsule. Other ingredients which may be present are talc, starch, cellulose, alkali-soluble water-insoluble cellulose ethers and higher fatty acids, such as oleic, palmitic and stearic acids. In one example, sodium penicillin is added to aqueous gelatine of pH 5 to 7 and temperature 50‹ C., the mixture cooled, dehydrated, the gel so formed treated with formaldehyde and the mass coated first with aniline formaldehyde resin and then with a 12 per cent free carboxy methyl cellulose ether.
