Abstract
2-Benzyl-4-methoxymethylene-5-oxazolone is manufactured by reacting a -phenacetamido-b : b -dimethoxypropionic acid with an excess of acetic anhydride, concentrating under reduced pressure to remove acetic acid and non-reacted acetic anhydride, washing with petroleum ether to cause the residue to solidify, and recrystallizing and drying the product. If desired, the product is hydrolysed in an aqueous alkaline solution (e.g. aqueous caustic soda) to form an alkali metal salt of 2-benzyl-4-hydroxymethylene-5-oxazolone, or, by acidification of the alkaline solution produced (e.g. with hydrochloric acid), the free hydroxy compound. The various stages are illustrated by examples. Specification 585,089 is referred to. a - Phenacetamido - b : b - dimethoxypropionic acid is obtained by treating methyl a -formyl-a -phenacetamidoacetate with dry methanol and dry hydrogen chloride to produce methyl a -phenacetamido-b : b -dimethoxypropionate, and treating this successively with an alkali metal hydroxide (e.g. caustic soda) and an acid (e.g. sulphuric acid) to form the free acid. Methyl a -formyl-a -phenacetamidoacetate is prepared by treating phenacetamidoacetic acid methyl ester with methyl formate in dry benzene in the presence of sodium methoxide and treating the product with dilute sulphuric acid. The Specification as open to inspection under Sect. 91 comprises also examples of the preparation of: (a) the dinitrophenylhydrazone of methyl a -formylphenaceturate by treating phenacetylglycine methyl ester with methyl formate in ether in the presence of sodium methoxide and treating the product with a hydrochloric acid solution of 2 : 4-dinitrophenylhydrazine; (b) the benzylamide of a -phenacetamido-b : b -dimethoxypropionic acid by heating the methyl ester of the acid with benzylamine; (c) b -methoxy-a -phenacetamidoacrylic acid by heating b : b -dimethoxy-a -phenacetamidopropionic acid with acetic anhydride, sodium acetate and piperidine; (d) 2-benzyl-4-benzyl-aminomethylene-5-oxazolone by treating 2-benzoyl - 4 - methoxymethyleneoxazolone with benzylamine in ether; (e) penaldic amide-2 : 4-dinitrophenylhydrazone by shaking 2-benzyl-4-methoxymethylene-5-oxazolone with aqueous ammonia and treating the product with 2 : 4-dinitrophenylhydrazine. This subject-matter does not appear in the Specification as accepted.
