Abstract
Histamine is removed from streptomycin by adsorbing said substances from their aqueous solutions on to an inorganic base exchange material and removing the streptomycin by selective elution with a dilute aqueous solution of a material capable of displacing the streptomycin therefrom by base exchange such as alkali metal and ammonium salts or hydroxides. Preferred base exchange materials are the natural or synthetic alkali metal or alkaline earth metal alumino-silicates especially sodium alumino-silicates. Preferred eluants are aqueous solutions of sodium chloride of a concentration of 1-8 per cent, preferably 3-5 per cent. When solutions of a concentration of more than 4 per cent are used for regenerating the base exchange material the resulting solution remains contaminated with histamine which may be removed by treatment with fresh base exchange material. Histidine present in the original streptomycin solution may be removed from the base exchange material by treatment with very dilute sodium chloride solution, e.g. 0.5 to 1.0 per cent or a 0.5 to 2 per cent pyridine solution. The histamine may be recovered by its salt with 2 : 4-dinitro-1-naphthol-7-sulphonic acid. In examples: (1) streptomycin is purified by adsorption on a synthetic sodium alumino-silicate and elution with 1 per cent sodium chloride; (2) streptomycin is freed from histidine and histamine by adsorption on synthetic sodium aluminosilicate, washing with 1 per cent aqueous pyridine, elution with 5 per cent sodium chloride and passing the resultant solution through a fresh bed of ion-exchange material to remove the remaining histamine.
