Abstract

12-Keto-steroid carboxylic acids are prepared by reacting a 3 : 12-dihydroxy substituted in the cholanic acid or a lower homologue thereof, with an anhydride or an acid chloride of a dibasic carboxylic acid, and oxidizing the partial ester so formed. If desired, the half ester may finally be saponified by treatment with alkali. The preferred dicarboxylic acids are succinic and phthalic acids. In examples: (1) Desoxycholic acid is treated with succinic acid in pyridine solution to give the 3-acid succinate which is then oxidized with cold aqueous chromic acid. Alternatively, the oxidation may be carried out with nitric acid. (2) The 3-acid succinate of nor-desoxycholic acid is prepared as in example (1) and oxidized with cold aqueous chromic acid, and finally saponified with aqueous alkali. (3) The 3-acid succinate of bisnor-desoxycholic acid is prepared in example (1) and is oxidized and saponified as in example (2). (4) Etio-desoxycholic acid is converted into its acid succinate, oxidized and saponified as in example (2). The Specification as open to inspection under Sect. 91 includes also the use as a starting material of any 3 : 12-dihydroxy-steroid compound substituted in the 17-position by a carboxylic acid group or a hydrocarbon chain terminated by a carboxylic acid group. This subject-matter does not appear in the Specification as accepted.
