Abstract

The antirachitically active component is separated from irradiated 7-dehydrocholesterol by steps comprising esterification of the irradiated product, separating the mixed esters by differential solution, saponifying the portion containing the active ingredient and at any stage, i.e. before esterification, in the esterified form or after saponification, effecting a purification by the method of chromatographic adsorption. The crude irradiation products may be first purified by treatment with maleic anhydride or its homologues (e.g. citraconic anhydride) to produce the ester which is separated and saponified. Suitable esterifying groups for combination with the vitamin, (referred to as vitamin D3) in order to form relatively sparingly soluble esters are allophanic acid (the ester is prepared by leading cyanic acid gas into the solution of the material) and 3-5-dinitrobenzoic acid or its 4-methyl derivative. Detailed descriptions of preparation of the vitamin are described in an example and in the schedule to a sample filed under Sect. 2 (5). The Specification as open to inspection under Sect. 91 includes examples and the schedule to a sample in which the step of chromatographic adsorption is not included. This subject-matter does not appear in the Specification as accepted.
