Abstract

328,347. Wellcome Foundation, Ltd., Smith, S., and Ward, V. J. Feb. 5, 1929. Medicines for internal use.-The active principles of the posterior lobe of the pituitary gland are purified and fractionally separated by employ. ing phenol or cresol, or solutions thereof in alcohol or like solvent as an extracting medium for the glands, or as a solvent for the material salted out from an aqueous extract of the gland by means of ammonium sulphate, or for active protein matter obtained by adding picric, picrolonic or flavianic acid to an aqueous pituitary extract, grinding the precipitate so produced with alcoholic hydrochloric acid and pouring the turbid mixture into acetone to obtain a precipitate. The solution of active materials obtained by any of the above methods is poured into a mixture of ether and light petroleum to obtain a precipitate having relatively great oxytocic and pressor activity. This precipitate is again dissolved in phenol, cresol, or an alcoholic solution thereof and the solution is treated first with ether to give a precipitate having great pressor activity, and then, after saturation with water, with light petroleum to throw down an aqueous layer having substantially only oxytocic activity.
