Abstract

Uric acid as the monovalent urate is a potent endogenous antioxidant protecting against free radical damage in man and other animals. Aging of human skin characterised by wrinkling results from free radical damage to vital tissues beneath the stratum corneum. Synthetic urate spheres formed in a cation rich aqueous solution at ambient temperature are extracted in ethanol and selectively filtered for 0.5žm or less; following air-drying being incorporated in a simple ointment base for skin application. Negatively charged particles of 0.5žm or less, as represented by these selected synthetic urate spheres, can permeate the barrier of the stratum corneum enabling free radical scavenging. These spheres show stability in various physiological media and added to the low solubility of urate, ensures prolonged local tissue activity; also precluding the risk of systemic absorption causing gout, although excessive application may cause local irritation.

The Dcsciiption.
Title: Anti-aithi skin cream.
Background:
Aging of skin chaiacterised by wnnlding is a direct result of flee radical damage, associated with the decrease in antioxidant molecules in the skin of old animals (Kohen et a! 2004). Uric acid is recognised as a potent natural antioxidant in living tissues (Becker 1993), with a specific role as a free radical scavenger. Spherulitic uric acid (urate spheres') form in aqueous solution under high cation concentrations (Mc Nabb 1980), these sphere are size modulated (<50pin) and do not coalesce in alcohol or dry powder form, giving even dispersal characteristics a major advantage over simple ground samples. IJrate spheres having a negative charge characterislic and if small enough (05i1m) could penetrate the intact skin (Kohli and Alpar 2004). where they would supplement age depleted anlioxidants. EJiate spheres made by this method show remarkable stability in various physiological environments and are well tolerated in open skin wounds allowing for the possibility of tissue incorporation (unpublished own work 2005).
Essential features: Microscopic spherulitic urate as a skin absorbable anti-aging agent.
Method: Uric acid is converted into spherulitic urate under saturated acpieous conditions using concentrated sodium hydroxide at room temperature. The aqueous phase is replaced by absolute ethanol (98%), which preserves these uratc spheres. Selective size filtration is used to produce 0.5tm spheres followed by evaporative drying giving a fine powder of urate spheres for inclusion in a simple skin applicable ointment base like soft paraffin and lanolin.

Claims (2)

CLAIMS.
1. A cream composed of size selected synthetic urate spheres in an ointment base for intact skin application to counteract the effect of aging from free radical damage.
2. Size selected synthetic urate spheres claimed in Claim I made by mixing aqueous solutions of saturate uric acid and concentrated sodium hydroxide replacing the aqueous phase with ethanol, filtering to O.5j.tm in size, dried and incorporated in a simple ointment base for skin application.
