Abstract

1479964 2,3-Methyleneprostaglandins SANDOZ Ltd 8 July 1974 [9 July 1973 4 May 1974] 30268/74 Heading C2C [Also in Division C3] The invention comprises optically active 2,3- methyleneprostaglandins of the formulµ or racemates thereof, wherein R 1 is H, C 1-8 alkyl, C 3-10 cycloalkyl or C 7-12 aralkyl, R 2 and R 3 each are H or C 1-4 alkyl, R 4 is 2-tetrahydropyranyl, R 6 is H, C 1-8 alkyl, C 3-10 cycloalkyl or C 7-12 aralkyl, and D is one of the four carbocyclic radicals IIa to IId and A is a carbocyclic radical of Formula IIa or IIb above or of the formula IIe or IIf and salts thereof when R 1 is H and their preparation. The compounds of Formula I above are prepared by hydrolysing the corresponding compound of Formula III, or when D is a radical of the Formula IIb by the acidic dehydration of compound of Formula I above in which D is a carbocyclic radical of the Formula IId. The esters and acids of the compound of Formula I above are obtained by esterification of the corresponding free acids and hydrolysis of the appropriate esters respectively. Compounds of the Formula III above in which A is a carbocyclic radical of the Formula IIa, IIb or IIf are obtained by oxidizing the corresponding compounds of Formula III in which A is a carbocyclic radical of the Formula IIe above or IIg or IIh Compound of Formula III in which A is a carbocyclic radical of the Formula IIe are prepared by reacting compounds of the formula E-CH=CH-CH(OR 4 )-C(R 2 )R 3 -(CH 2 ) 3 CH 3 wherein E is a radical of the Formula Vc with phosphonium halides of the Formula VI wherein X is Cl, BrI and R 5 is naphthyl or phenyl optionally substituted by alkyl, or alkyl. Pharmaceutical compositions contain the above 2,3-methylene prostaglandins of the Formula I above in association with pharmaceutical carriers or diluents. The compounds possess prostaglandin-like activity.
