Abstract

1472767 Preparation of cinnoline derivatives IMPERIAL CHEMICAL INDUSTRIES Ltd 23 Sept 1975 [7 Nov 1974] 48206/74 Heading C2C Cinnoline derivatives having the general formula wherein R<SP>1</SP> is hydroxy or C 1-6 alkoxy, are prepared by first diazotizing a compound of the formula either (a) by means of nitrous acid in an aqueous acidic medium at approximately 0‹ C., or (b) by means of a C 1-6 alkyl nitrite or phenyl nitrite, and then reacting the resulting diazonium compound with benzene. The nitrous acid may be generated in situ from an alkali metal nitrite and an inorganic acid and the preferred alkyl nitrite is amyl nitrite. The starting material of formula III may be prepared by catalytic hydrogenation of the corresponding alkyl 4-hydroxy nitrocinnolin-3-yl carboxylate which in turn may be prepared by esterification of the corresponding 4 - hydroxy - nitrocinnolin - 3 - yl carboxylic acid. The compounds of formula I are active, as inhibitors of the effects following the combination of reagin-like antibodies and their antigens.
