1317257 Thiazole derivatives LILLY INDUSTRIES Ltd 6 July 1970 [15 July 1969] 54456/72 Divided out of 1317256 Heading C2C Novel compounds of Formula I (in which Y is O or S, n is 0 or 1 and R is H or C 1-6 alkyl) are prepared (a) by condensing a compound of Formula II with a 1-halomethylglyoxal or with an acetal thereof, or with a compound of the formula Hal.CH 2 .CO.CO.R (where Hal is halogen and R is C 1-6 alkyl), or (b) by reacting a compound of Formula III with an alkali metal salt of an acid of formula R<SP>111</SP>COOH (where R<SP>111</SP> is an optionally substituted hydrocarbon radical) to replace the halogen atom by the radical O.CO.R<SP>111</SP>, hydrolysing the resulting ester in the presence of a mineral acid and oxidizing the resulting alcohol to the aldehyde or ketone. The compounds I in which R is H may also be prepared by (i) treating a compound of the Formula IV with p-nitroso dimethyl aniline in the presence of a catalyst and subjecting the resulting N-(pdimethylaminophenyl) - 2 - substituted - 4 - thiazolyl nitrone to acid hydrolysis, or (ii) treating a compound of Formula V with dilute acetic acid. Compounds of Formulae IV and V are prepared by reacting a compound of Formula III (in which R is H) with hexamine or pyridine. 2 - (5 - Nitro - 2 - furyl) - 4 - chloromethylthiazole is prepared by reacting 5-nitro-2-thiocarbamyl-furan with 1,3-dichloro-acetone. The semicarbazone of 2-(5-nitro-2-thienyl)-4- formyl-thiazole and the 2,4-dinitrophenyl hydrazone of 2 - (5 - nitro - 2 - furyl) - 4 - formylthiazole are prepared.
