Abstract

1472325 Pyrazolo[4,3-c]pyridine derivatives E R SQUIBB & SONS Inc 13 June 1974 [28 June 1973] 26273/74 Heading C2C The invention comprises compounds of formula and their N-oxides and acid addition salts, wherein R and R<SP>3</SP> are each Ph, thienyl or pyridyl; R<SP>1</SP> is H, C 1-8 alkyl (possibly HO- or Phsubstituted) or C 2-9 alkanoyl; R<SP>2</SP> is B-C 1-8 -alkyl where B is mono- or di-(C 1-8 -alkyl)amino, piperidino, pyrrolidino, morpholino or N-(C 1-8 - alkyl or HOC 2 H 4 )-piperazino; and said Ph groups may be substituted by Cl, F, CF 3 or C 1-8 alkyl or alkoxy. These compounds are prepared by reacting (1) R<SP>2</SP>NHNH 2 with the appropriate 3,5-bis(arylmethylene)-1-R<SP>1</SP>-4-piperidone, followed if desired by N-alkanoylation or (2) an analogue of I where R<SP>2</SP> is tosyloxyalkyl with the base BH. Therapeutic compositions having C.N.S. depressant activity comprise compounds of the above formula, and may be administered orally (preferably), rectally, intraperitoneally, subcutaneously, intramuscularly or intravenously.
