Abstract

1447338 Pyridoxine mono-esters SOC D'ETUDES DE PRODUITS CHIMIQUES 16 April 1975 15552/75 Addition to 1286161 Heading C2V A process for the preparation of a salt of a pyridoxine mono-ester of formula in which A and A<SP>1</SP> are alkylene radicals or divalent amino-substituted aliphatic hydrocarbon radicals; R<SP>1</SP> and R<SP>2</SP> are alkyl radicals or R1 and R<SP>2</SP> form, together with the nitrogen atom, a heterocyclic ring; Ac represents a pharmaceutically acceptable acid; and n is 1, 2 or 3, comprises blocking the two hydroxyl groups in the 3- and 4-positions of pyridoxine, reacting the compound thus obtained with an acid anhydride of formula converting the resulting acid monoester intermediate into its acid chloride, reacting the acid chloride with a tertiary amine of the general formula HO-A<SP>1</SP>-NR<SP>1</SP>R<SP>2</SP>, reacting the resulting compound with a pharmaceutically acceptable acid to give a pharmaceutically acceptable acid addition salt and removing the group blocking the two hydroxyl groups at the 3- and 4-positions by heating the mono-ester. In examples pyridoxine is reacted with acetone to block the two hydroxyl groups and the (4,3,0)- isopropylidene pyridoxine so obtained is reacted with succinic anhydride and with glutaric anhydride, the products then being reacted with thionyl chloride to obtain the acid chlorides which are reacted with dimethyl aminoethanol and with 4-methyl-1-piperazinyl ethanol; the resulting compounds are reacted with maleic acid, with N-acetyl aspartic acid and with maleic anhydride and the blocking groups removed by heating.
