Abstract

1431940 Copolymer carriers for biologically active substances ROHM GmbH 30 July 1973 [29 July 1972] 36218/73 Headings C3H and C3P Copolymers in the form of particulate, cross-linked, water-swellable, spherical beads suitable for use as carriers for biologically active substances containing at least one primary amino or hydroxyl group, are prepared by copolymerizing a dispersion in a non-polar organic medium of a monomer mixture comprising:- (a) at least one radically copolymerizable monomer containing a group capable of reacting with the primary amino or hydroxyl group or groups of the biologically active substance with the formation of a covalent bond; (b) at least one monomer containing at least two radically copolymerizable carbon-carbon double bonds; (c) one or more radically copolymerizable water-soluble monomers, i.e. those which, at 20‹ C., give an at least 10% by weight aqueous solution; and (d) if desired, one or more radically copolymerizable water-insoluble monomers; component (b) amounting to 0À2 to 5 mol per cent of the monomer mixture, and the amounts of components (a), (b) and (c) being selected to provide a total amount of 50 to 100 mol per cent of the monomer mixture and to provide beads of the resulting copolymer which are capable of swelling in water to 5 to 100 times their original volume; and subsequently separating the resulting particulate copolymer from the organic medium. In the examples acrylamide, methacrylic anhydride, ethylene glycol dimethacrylate, acrylic acid, triethylene glycol dimethacrylate, acrylic anhydride, methacrylamide, acryloyl N- hydroxy-succinimide and glycidyl acrylate are copolymerized in the presence of n-heptane, perchloroethylene, benzoyl peroxide, dimethylformamide, n-butyl methacrylate/methacryloylcholine hydrochloride copolymer as emulsifier, dimethylaniline, formamide, an aliphatic oil, a stabilizer and 4,4-azobis-(4-cyano-valeric acid). The resulting polymer beads are reacted with trypsin, chymotrypsin, pancreatin, protease of Streptomyces griseus, ribonuclease A, desoxyribonuclease I, insulin, glucagon, hypertensin, trypsin inhibitors from pancrease, tyramine and antibiotic ampicillin.
