Abstract

1409539 Adenosine-5-carboxylic acid esters ABBOTT LABORATORIES 8 Sept 1972 [10 Sept 1971] 41943/72 Addition to 1408201 which is addition to 1350671 Heading C2C Compounds I in which R 1 is C 1-6 alkyl, or cyclopropylmethyl and R 2 and R 3 are both hydrogen, or when R 1 is C 1-6 alkyl R 2 and R 3 may be taken together to form an isoalkylidene group are prepared by either (1) reacting an appropriate adenosine-5- carboxylic acid chloride with an alcohol R 1 -OH and thereafter treating the product with an acid acceptor in the liquid phase or (2) adding an appropriate adenosine-5-carboxylic acid to an alcohol R 1 -OH and treating the resulting solution or dispersion with thionyl chloride and treating the resultant product with an acid acceptor in the liquid phase and thereafter if desired hydrolysing a product obtained in which R 2 and R 3 are isoalkylidene to that in which R 2 and R 3 are hydrogen. Compounds I are also obtained by reacting thallous adenosine-5- carboxyl with R 1 -X in which X is chlorine, bromine or iodine. Compounds I in which R 1 is C 1-6 alkyl and R 2 and R 3 are hydrogen are also obtained by heating a solution of a halo C 1-6 alkyl adenosine-5-carbosylate, in an inert solvent with an alkanol of 1 to 6 C atoms. Compounds I are blood-pressure lowering agents. Reference has been directed by the Comptroller to Specification 1,295,229.
