Abstract

1389822 Substituted benzopyrans BEECHAM GROUP Ltd 26 April 1972 [11 May 1971] 14226/71 Heading C2C Novel compounds of the Formula (II) wherein R 1 and R 2 which may be the same or different are each a C 1-6 alkyl group, R 3 is a non-aromatic C 1-12 hydrocarbon group, R 4 is an optionally substituted phenyl group, and X is a hydrogen atom, a salting ion or an acyl or inorganic ester group, are obtained (a) by reaction of a chromanone of the Formula (IV) or salts thereof with an organometallic compound of formula R 4 Y, where Y is an alkali metal or magnesium-halogen group, treating with a proton source to liberate a compound of Formula (V) and allowing or causing the dehydration of this compound to yield (II), or (b) by reaction of an organometallic compound R 1 Y where Y is an alkali metal or magnesium halide upon a coumarin (VII) treating the resulting intermediate with a proton source to yield a compound of the Formula (VIII) and causing or allowing the dehydration of VIII to yield II, and therafter, if required in either case, converting one group R 4 or X to another group R 4 or X by known methods. Intermediates (VII), may be obtained by reaction of a #-benzoyl acetate R 4 .CO.CH 2 .CO 2 R 5 (where R 5 is an ester forming group) with a 3- R3-substituted resorcinol in the presence of a condensing agent. Pharmaceutical compositions, having action on the central nervous system, comprise compounds of Formula (II) together with a pharmaceutical carrier in forms for oral administration.
