Abstract

1367928 2-Chloro-dexamethasone CIBAGEIGY AG 10 Dec 1971 [10 Dec 1970] 57465/71 Heading C2U The novel #<SP>1,4</SP> - 2 - chloro - 9α - fluoro - 16α- methyl - 11#,17α,21 - trihydroxy - 3,20 - dioxopregnadiene is prepared (i) by addition of chlorine to the #<SP>1</SP>-double bond of dexamethasone or a hydroxy-protected derivative thereof with subsequent splitting off of HCl from the resulting 1,2-dichloro compound, or (ii) by treatment of #<SP>1,4</SP> - 2 - chloro - 9#,11# - epoxy - 16α - methyl - 17α-21 - dihydroxy - 3,20 - dioxo - pregnadiene or a 21-protected derivative thereof with HF or a suitable derivative thereof, with removal of protection from hydroxy groups, e.g. hydrolysis of esters, when required. A specified starting material is dexamethasone 11-trifluoroacetate 21-acetate (prepared by trifluoroacetylation of dexamethasone to give dexamethasone 11,21-bistrifluoroacetate, hydrolysis of this to dexamethasone 11-trifluoroacetate and acetylation of this) which is finally hydrolysed in 2 stages, the 11-acyloxy group being hydrolysed first and then the 21-acetoxy group. The novel steroid is an anti-inflammatory agent, and it may be made up into pharmaceutical compositions with suitable carriers.
