Abstract

1367576 Tetrasubstituted imidazolidines AMERICAN CYANAMID CO 10 Oct 1972 [21 Oct 1971] 46735/72 Heading C2C [Also in Division A5] The invention comprises compounds having the general formula wherein X is a hydrogen or chlorine atom; and Y and Z are each NH, NCH 3 or O; with the proviso that both Y and Z are not O in the same compound, and pharmaceutically acceptable salts thereof. They may be prepared by the consecutive steps of (a) reacting an aniline of the formula with an acetic acid derivative of the formula to produce an intermediate of the formula (b) cyclizing Formula IV to form an imidazolidine of Formula I wherein B and D are NHR, NRCN or B and D not being the same and when B is D may also be OR, SR or NHR, A is a leaving group; B<SP>1</SP> is cyano, hydrogen or when B<SP>1</SP> is hydrogen, D can also be OCN; provided that when D is NHR, B<SP>1</SP> CN or and when D is NRCN or B<SP>1</SP> is hydrogen; Z<SP>1</SP> is NH 2 , OCH 3 , OH, SCH 3 , or NHCH 3 ; and R is CH 3 or hydrogen in which case the imidazolidine is methylated after step (b). The compounds of the invention may be used to antagonize renal aldosteronic action and induce diuresis alone or in admixture with conventional carriers and/or other diuretic agents. They may be administered parenterally in solution in a polyhydric aliphatic alcohol which may also contain preservatives to prevent bacterial and fungal contamination, and antioxidants; and orally in the form of tablets, troches, capsules, elixirs, suspensions, syrups, wafers and chewing gum.
