Abstract
1,265,051. Perfluoroalkyl aralkylamines. MERCK & CO. Inc. 10 March, 1969 [13 March, 1968; 17 Feb., 1969], No. 12489/69. Heading C2C. Novel compounds of the Formula (I<SP>1</SP>) wherein n is 2 or 3, A<SP>11</SP> is a substituted or unsubstituted carbocyclic or heterocyclic ring of 5 or 6 atoms; B is an amino substituted straight or branched aliphatic side chain containing from 1 to 8 carbon atoms, and the phenyl ring optionally contains a substituent in addition to B and A<SP>11</SP>-(CF 2 ) n , may be obtained (a) by reducing a compound of Formula (III) to form a compound (I<SP>1</SP>) in which B is -CH 2 NH 2 preferably using LiAlH 4 . In preparing higher homologues in which B is -(CH 2 ) m NH 2 and m is 2, 3 or 4, the intermediate phenylperfluoroalkylaryl nitrile is converted as described in Specification 1,265,054 to the corresponding nitrile containing the group B= -(CH 2 ) m-1 CN which is reduced to the alkylamine. The amine may be N-alkylated to the corresponding N- alkylaminoalkyl or N,N-dialkylaminoalkyl compounds, e.g. by reacting the primary amine with the halide or anhydride of a C 2-5 aliphatic acid to form the N-alkanoylamide and reducing the latter (the tertiary amine being obtained by repeating this process on the secondary amine). Compounds (I<SP>1</SP>) in which B is -(CH 2 ) m NR 1 R 2 , and NR 1 R 2 is 1-pyrrolidinyl, 1-piperidyl, 4- morpholinyl, 4 - thiomorpholinyl or 1 - (C 1-5 alkyl) - 4 - piperazinyl are formed by condensing (I<SP>1</SP>) where B is -CH 2 NH 2 with an appropriate α,α-dihalo compound. The products may also be obtained from a compound (I<SP>1</SP>) in which B is halogen by converting B to -MgHal, treating with CO 2 to form B = -COOH, reducing with LiAlH 4 to form B = -CH 2 OH, converting to the benzyl halide and reacting with ammonia or an appropriate amine. Compound III may also be reacted with ethylenediamine to form a compound (I<SP>1</SP>) in which B is imidazoline. Pharmaceutical compositions useful in treatment or prevention of cardiac arrhythmias comprise a compound of Formula I<SP>1</SP> above and an inert non-toxic diluent, carrier, coating or excipient in forms suitable for oral or parenteral administration.
