1364314 Indole derivatives JOHN WYETH & BRO Ltd 10 Feb 1972 [11 Feb 1971] 4458/71 Addition to 1218570 Heading C2C The invention comprises novel indole derivatives of the Formula VI wherein R<SP>1</SP> is H, C 1 -C 6 alkyl, C 7 -C 10 aralkyl, or aroyl; R<SP>2</SP> is H, C 1 -C 6 alkyl or aryl; R<SP>3</SP> is H, halogen, OH, C 1 -C 6 alkyl or C 1 -C 6 alkoxy; R<SP>4</SP> is H, halogen or C 1 -C 6 alkyl; R<SP>5</SP> is aryl (including heteroaryl), C 1 -C 6 alkoxy, aryloxy, C 7 -C 10 aralkyl, C 7 -C 10 aralkyloxy or diaryl- C 1 -C 6 alkyl; A is C 1 -C 4 hydroxyalkylene; and the formula is a pyridinium, 1,2,3,6-tetrahydropyridine or piperidine, substituted by NHOOR<SP>5</SP> and R<SP>4</SP>, and the pharmaceutically acceptable acid addition and quaternary ammonium salts of the tetrahydropyridine and piperidine compounds. The compounds are prepared by one of the following methods:- (a) selectively reducing corresponding compounds of the Formula VI above in which A is a mono- or diketoalkylene radical. (b) Acylating the corresponding compounds of the Formula VI above in which the NHCOR<SP>5</SP> radical is replaced by an NH 2 radical with reactive derivatives of acids of the formula R<SP>5</SP>COOH. (c) Selectively reducing the corresponding pyridinium compounds with alkali metal borohydrides to the tetrahydropyridine compounds. (d) Reducing the corresponding pyridinium or tetrahydropyridine compounds to the corresponding piperidine compounds. (e) Alkylating, aralkylating or aroylating the corresponding compounds of the Formula VI above in which R<SP>1</SP> is H. 3 - [1 - Oxo - 2 - (4 - benramidopiperid - 1 - yl) ethyl]-indole is obtained by reacting 3-indolyl bromomethyl ketone with 4-benzamidopiperidine. Pharmaceutical compositions, suitable for oral or parenteral administration, contain the above novel compounds or pharmaceutically acceptable acid addition or quaternary ammonium salts thereof and pharmaceutically acceptable carriers. The compounds possess hypotensive and/or anti-histamine activity.
