1399608 Derivatives of 3-(piperidinoalkyl)- indoles JOHN WYETH & BRO Ltd 6 Nov 1972 [19 Nov 1971] 53782/71 Addition to 1218570 Heading C2C The invention comprises compounds of formula wherein R<SP>1</SP> is H, C 1-6 alkyl, C 7-10 aralkyl or aroyl; R<SP>2</SP> is H, C 1-6 alkyl or aryl; R<SP>3</SP> is H, halogen, OH or C 1-6 alkyl or alkoxy; R<SP>4</SP> is H, halogen or C 1-6 alkyl; R<SP>5</SP> is aryl (possibly heterocyclic), C 1-6 alkoxy, aryloxy, C 7-10 aralkyl or aralkoxy, diaryl-C 1-6 -alkyl or C 5-7 cycloalkyl; R<SP>6</SP> is an organic quaternizing group removable under mild conditions, e.g. by hydrogenolysis; X is an anion; A is C 1-4 alkylene (possibly substituted by OH or 1-2 oxo groups which may be ketalized); Z is oxo when the bond represented by the broken line is absent, or is oxo or H 2 when that bond is present. These compounds, which are prepared by reacting together the appropriate 3-X-A-indole and 1-R<SP>6</SP>-piperidine, may be hydrogenolysed to give the corresponding unquaternized compounds (in which R<SP>6</SP> and X are absent). Therapeutic compositions for oral or parenteral administration comprise compounds of the above formula, which act on the cardiovascular system (e.g. hypotensive and/or antihypertensive activity).
