Abstract

1355866 Dibenzoxazepine derivatives WANDER Ltd 2 Aug 1971 [6 Aug 1970 1 June 1971] 36235/71 Heading C2C Novel compounds I (in which A is C 1-3 alkylene and R 1 is C 3-8 aliphatic hydrocarbyl) and their acid addition salts, are prepared by (a) reacting an acid R 1 COOH or a salt, acid halide or acid anhydride thereof, with a compound III (in which X is halogen, tosyl, OH, OMe, where Me is a metal, or (b) reacting a compound V with a compound IV (in which Y is halogen, C 1-4 alkoxy, C 1-4 alkylthio, SH or p-nitrobenzylthio) or (c) reacting the compound VI with a compound Y-A-O-CO-R 1 . 2 - Trifluoromethylsulphonyl - 11 - (1 - piperazinyl or 4-#-hydroxyethyl-1-piperazinyl)-dibenz- [b,f][1,4]oxazepine is prepared by reacting 2- 2 - trifluoromethylsulphonyl - 11 - chlorodibenz- [b,f][1,4]oxazepine with piperazine or 4-#- hydroxyethylpiperazine. 2 - Trifluoromethylsulphonyl - 11 - chlorodibenz[b,f][1,4]oxazepine is prepared by reacting 2-nitro-4<SP>1</SP>-methylthio-diphenyl oxide with chlorine to give 2-nitro-4<SP>1</SP>-trichloromethylthio-diphenyl oxide, reacting this with SbF 3 to give 2- nitro-4<SP>1</SP>-trifluoromethylthiodiphenyl oxide, hydrogenating this to give 2-amino-4<SP>1</SP>-trifluoromethylthio-diphenyl oxide, reacting this with phosgene to give 2-isocyanato-4<SP>1</SP>-trifluoromethylthiodiphenyl oxide, reacting this with phosphorus oxychloride and phosphorus pentoxide to give 2 - trifluoromethylthio - 10,11 - dihydro - 11 - oxodibenz[b,f][1,4]oxazepine, reacting this with H 2 O 2 to give 2-trifluoromethylsulphonyl-10,11- dihydro-11-oxo-dibenz[b,f][1,4]oxazepine and reacting this with POCl 3 . Compounds V are prepared by acylating a compound XVII and subsequently debenzylating the reaction product. Pharmaceutical compositions having CNS, neuroleptic and antiemetic activity, for parenteral administration, comprise a compound I, or a pharmaceutically acceptable acid addition salt thereof, together with a suitable carrier.
