1321691 Derivatives of 4-hydroxy-1-phenethylpiperidine SANDOZ Ltd 16 June 1970 [8 July 1969 (2) 5 Aug 1969 12 Nov 1969 5 Dec 1969 5 March 1970 21 April 1970 (3)] 29233/70 Heading C2C The invention comprises compounds of formula and their acid addition salts, wherein R 1 is H or C 1-4 alkyl and R 2 is H, F, Cl, Br, C 1-4 alkyl, alkoxy or alkylthio, CN, NH 2 , CF 3 or NHCO- alkyl (C 1-4 ), provided that when R 2 is H or o-Cl, R 1 is C 1-4 alkyl. These compounds are prepared by (1) phenethylation (except where R 2 = o-NH 2 ) of a 4-hydroxy-4-(2-oxo-5-R 1 -3- tetrahydrofuryl)piperidine (which is made by hydrogenolysis of the N-benzyl derivative), in an inert solvent in presence of an acid-binding agent; (2) reaction of a 1-phenethyl-4-piperidone (except where R 2 = CN or NHCO-alkyl made by acid hydrolysis of the corresponding ethylene ketal) with a 2-oxo-5-R 1 -tetrahydrofuran, in an inert solvent in presence of a boric condensation agent, followed by hydrolysis; (3) (where R 2 = NH 2 ) reduction of the corresponding nitrocompound. Therapeutic compositions preferably in a form for oral administration comprise compounds of the above formula, which have analgesic properties.
