PFIZER Ltd 22 Aug 1969 [23 Aug 1968] 40412/ 68 Heading A5B Oral dosage units containing one or more pharmaceutically active ingredients comprising cores having an outer layer thereon consisting of particles of pharmaceutically active material or other material of pharmaceutical utility in a water-soluble matrix are prepared by forming the outer layer by spraying the cores with a suspension of the particles in a liquid medium comprising a water-soluble binding agent and a water-soluble cellulosic film-forming agent dissolved in a mixture of water and a watermiscible pharmaceutically acceptable organic liquid having a boiling point below 90‹C., and evaporating the solvent from the sprayed cores. The organic liquid may be acetone, ethanol or isopropanol; the binding agent may be sucrose, gelatine or gum acacia and the cellulosic film-forming agent may be hydroxypropyl methyl, methyl or sodium carboxymethyl cellulose. The particles deposited may form at least 10% by weight of the dosage unit. The cores may be sprayed with a suspension of a pharmaceutically active ingredient such as paracetamol, the core containing aspirin. The particles of material of pharmaceutical utility may comprise a pigment such as titanium dioxide. The core may be separated from the outer layer by an enteric layer of cellulose acetate phthalate.
