Abstract
1,238,971. Histidine containing peptides. FARBWERKE HOECHST A.G. 3 March, 1969 [2 March, 1968], No. 11239/69. Heading C2C. Peptides containing at least one histidine unit are prepared by treating a peptide containing a substituted histidine unit of the formula (wherein R 1 and R 2 each represent an alkyl, aralkyl or aryl group and in which the groups represented by R 1 and R 2 may be the same or different, or in which NR 1 R 2 represents a heterocyclic ring) with (a) NH 3 , a primary or secondary amine, a hydrazine or a hydrazine derivative having at least one -NH-group or (b) an alkali metal or alkaline earth metal hydroxide or (c) an alkali metal or alkaline earth metal and liquid NH 3 or (d) an alkali metal alcoholate. The protected histidine containing peptides are prepared by reacting an N<SP>α</SP>-acyl histidine ester with a substituted carbamoyl chloride in the presence of an acid binding agent and then building the peptide by known methods or by acylating the N<SP>Im</SP> atom in an otherwise fully protected histidine-containing peptide with a carbamoyl halide. Protecting groups other than the carbamoyl groups protecting the N<SP>Im</SP> atom of histidine may be removed by known means before or after removing the N<SP>Im</SP> protecting group. Z-His-OBzl and Z-Phe-His-OBzl are prepared as starting materials. Pharmaceutical compositions comprise a histidine peptide of the invention together with a suitable carrier.
