Abstract

[Also in Division C3] An enzyme system comprises amylo-1,6- glucosidase and glucoamylase and is able to saccharify thinned starch to dextrose under conditions which tend to inhibit the reversionary action of glucoamylase. The amylo-1,6-glucosidase may be produced by Aerobacter aerogenes and the glucoamylase by Aspergillus niger, Aspergillus phoenicis, or species of Clostridium, Mucor or Rhizopus. The starch to be saccharified may be thinned by partial acid hydrolysis, but is preferably thinned by the action of alphaamylase. In examples, corn starch is cooked, thinned by alpha-amylase, and saccharified with varying amounts of glucoamylase and amylo-1, 6-glucosidase and at varying conditions of temperature and pH. Over 99% conversion to dextrose was achieved at 50‹ C. and pH 5.8 to 6.2.
