Abstract
1,206,723. α-Thiolincosaminides. UPJOHN CO. 1 Feb., 1968 [13 Feb., 1967], No. 5184/68. Heading C2C. The invention comprises novel compounds of the general formulae and the acid addition salts thereof, wherein R 2 is C 1-4 alkyl; R 3 is H or an acyl, or a halo-, nitro, -hydroxy-, amino-, cyano-, or thiocyanosubstituted hydrocarbon carboxylic acid acyl radical, the acyl moiety having up to 18 C atoms, and X is CH 3 or C 2 H 5 . Compounds of formula (1) wherein R 2 and R 3 are C 1-4 alkyl or wherein R 3 = H and R 2 is C 1-4 alkyl and where X is CH 3 are obtained by the hydrogenation of an N, O-acetal of methyl α-thiolincosaminide to give N,N-dialkyl-methyl-α-thiolincosaminide and N-alkyl-methyl-α-thiolincosaminide. Compounds of formula (2) wherein R 2 = C 1-4 alkyl and X is CH 3 or C 2 H 5 are obtained by alkylating a compound of general formula with 1 mole of alkylating agent. Amongst the examples N-benzylmethylene-methyl-α-thiolincosaminide is hydrogenolyzed to give (a) N,N - dimethyl - methyl - α - thiolincosaminide and (b) N-methyl-methyl-α-thiolincosaminide and the latter is acylated with acetic anhydride to the penta-acetate.
