Abstract

1,199,862. Benzodiazepine derivatives preparation. F. HOFFMANN-LA ROCHE & CO. A.G. 5 Feb., 1968 [6 Feb., 1967], No. 5706/68. Heading C2C. Compounds of the general Formula (I) (A = phenyl, pyridyl, halophenyl; B=CO, CH 2 ; R, R<SP>1</SP> = halogen, CF 3 , NO 2 ; R<SP>2</SP> = C 1-7 alkyl, C 2-7 alkenyl; R<SP>3</SP> = C 1-7 alkyl; broken lines denote optional substituents) and their acid addition salts are prepared by treating a compound of the Formula (II) with a base (X = arylsulphonyl, C 1-7 alkanesulphonyl) or alkali metal C 1-7 alkoxide or hydride (X = C 1-7 alkanoyl) in the presence of an inert organic solvent, optionally followed by salt formation. When B=CO a compound of the Formula (III): may be obtained as an intermediate and can be converted, with or without isolation, to (I) by treatment with a base. The preparation of starting materials of Formula (II) is described.
