Abstract

1,195,854. 2-Oxazolecarboxylic acids and esters; pyridoxine. AJINOMOTO CO. Inc. 8 Aug., 1967 [8 Aug., 1966 (2); 3 Oct., 1966; 10 Jan., 1967], No. 36474/67. Headings C2C and C2V. Novel compounds of the general formula wherein R 1 is H or a carbo lower alkoxy group where the lower alkoxy group has 1 to 4 C atoms, R 2 is a C 1-4 alkyl and R 3 is H or a C 1-4 alkyl, are prepared by reacting a compound of the general formula with a dehydrating agent in the presence of an organic base. The dehydrating agent is preferably phosgene and the base used may be triethylamine. The esters may be converted to the acids by hydrolysing with an aqueous solution of an alkali metal hydroxide or alkaline earth metal hydroxide. In the Examples (1 and 8) the ethyl ester of alanine is condensed with diethyl oxalate to give ethoxalylalanine which is dehydrated using phosgene in triethylamine to yield 4-methyl-5-ethoxy-2-oxazolecarboxylic acid ethyl ester. The starting material may be obtained by reacting a compound of the general formula R 1 CH 2 CH(NH 2 )COOR 2 with an alkyl diester of oxalic acid, R 3 OOCCOOR 3 . Pyridine derivatives are obtained by further reaction of the novel compounds with dienophiles such as fumaronitrile, 2,5-dihydrofuran and 4,7-dihydro-1,3-dioxepine. 4 - Methyl - 5 - alkoxy oxazoles are obtained from the novel acids or their salts by decarboxylation. Pyridoxine hydrochloride is obtained by reacting 5-ethoxy-4-methyl-2- oxazolecarboxylic acid with 4,7-dihydro-1,3-dioxepine, followed by the addition of HCI.
