1,161,896. Thyroid gland polypeptides. I. MACINTYRE. 24 Aug., 1966 [26 Aug., 1965; 7 Jan., 1966; 11 July, 1966],. No. 37968/ 66. Heading A5B. A hypocalcaemic polypaptide is obtained from mammalian thyroid gland by extraction with aqueous acid, salt fractionation of the extract, and absorption on gel and elution therefrom in formic acid solution, followed by lyophilisation or evaporation to give the product. The process preferably includes a dialysis of the acid extract, and precipitation with trichloracetic acid at any stage subsequent to the salt fractionation. The preferred starting material is a defatted acetone powder of pigs' thyroid. The polypeptide has a molecular weight of about 2700, and contains most common L-amino acides excepting cystine. Pharmaceutical compositions comprise the polypeptide or its physiologically tolerable acid addition salts or complexes with a carrier, in a form suitable for internal or parenteral administration, and may also contain further unspecified active ingredients. The polypeptide may be reacted with bases or acids, or may be complexed with e.g. hydroxypolygelatin, polyvinyl pyrrolidone, Protamine, polyphloretin phosphate, phytic acid, polyglutamic acid, or suitable sparingly soluble metal compounds; such as Al, Mg, Co, or Zn phosphates, pyrophosphates or hydroxides, to provide, prolonged release forms.
