Abstract
1,160,151. Pharmaceutically active protein metal chelates. DIAGNOSTIC DATA Inc. 7 Oct., 1966 [8 Oct., 1965; 31 Aug., 1966], No. 44977/66. Heading A5B. Pharmaceutical compositions for treating inflammatory conditions and virus infections comprise a non-toxic carrier and a therapeutically active protein metal chelate of one or more divalent metals with a non-antigenic globulin type protein, free from other proteins and having a configuration which is at least 80% helical, less than 15% of the total amino acids being serine, threonine, proline, cysteine and isoleucine, and having more amino acids with free basic groups than those with free acidic groups, the chelate being water soluble at pH 1-11 except at pH 4-6. The preferred divalent metals are Mg, Fe, Zn, Cu and Ca; others exemplified are Mn, Al, Pb, Ni, Ag, and Cr; chelates of Na (monovalent), silicon and boron are also mentioned. The chelate has a molecular weight of about 28,500, an average analysis of: C-50.02, H - 7.92, 0 - 25.55, N - 15.26, S - 0.43, P - nil, ash - less than 1.0, and contains about 5% carbohydrate, about 2 gm. atoms of metal per molecule and 205 amino acid residues per molecule, including 1 cysteine, 3 methionine, 9 arginine, 15 histidine, 22 lysine, and 40 aspartic and glutamic residues, 13 of the latter being amides. The ratio of acidic to basic amino acids is 0.565. The chelates are prepared by extracting a suitable proteinaceous materials, (e.g. animal organs and tissue, including marine organisms; protein rich vegetable sources, fungi, bacteria and yeasts) with buffer solution of pH 1-4 or 6-11 containing the required divalent metal ions, and isolating the chelate by conventional protein purification procedures (e.g. solvent, salt and pH fractation, denaturation of more labile proteins, electrophoresis and gel filtration, at below 5‹C). Transchelation techniques are optionally employed to replace one chelated metal by another. The pharmaceutical compositions may be in forms suitable for injection, topical application ( e.g. solutions, creams, ointments, or salves, preferably with surfactants and/or skin penetrants), or oral administration, (as an enteric coated tablet). Compositions also containing anti-inflammatory steroids are maintioned. The compositions are said to stimulate interferon production.
