Abstract

1,160,291. Antibiotics MM54 and MM73. IMPERIAL CHEMICAL INDUSTRIES Ltd. 2 Jan., 1967 [31 Jan., 1966], No. 4182/66. Heading C2A. Antibiotics MM54 and MM73, also known as cytochalasins C and D, are produced by cultivating metarrhizium anisopliae IMI 98373 in an aqueous nutrient medium containing an assimilable source of carbon and nitrogen and inorganic salts. The antibiotics are extracted from the clarified fermentation broth at pH 7À0 with a solvent such as chloroform. The crude extract is dissolved in acetone, mixed with silica gel, the acetone removed, the acetone-free mixture added to a column of silica gel and eluted successively with benzene, benzene and chloroform 9: 1 and benzene and chloroform 4: 1 and the eluates are discarded. Further elution with 1:1 and 1: 3 benzene/chloroform and chloroform alone enables MM54 and MM73 to be recovered. Further purification is by recrystallization from acetone. The antibiotics have the following formulµ wherein the group in MM54 represents and in MM73 represents Consequently MM54 is 1-benzyl-3a,4,5,7a-tetrahydro - 5 - hydroxy - 6,7 - dimethyl - 3a,4 - (11 - acetoxy - 8 - hydroxy - 6,8 - dimethyl - 7 - oxo - cycloundeca - 3,9 - dieno)isoindoline - 3 - one. The antibiotics inhibit cytoplasmic cleavage and cell motility in tissue culture cells.
