Abstract

1292010 The antibiotic #-rhodomycin V FARBENFABRIKEN BAYER AG 19 April 1971 [18 March 1970] 26602/71 Heading C2A The new antibiotic #-rhodomycin V having the Formula I wherein R is a tetraglycoside radical consisting of 1 mol. 2-desoxy-fucose, 1 mol. rhodosamine and 2 mols. rhodinose, is prepared by aerobically cultivating a #-rhodomycin V-producing strain of Streptomyces purpurascens (e.g CBS. 296.71) and isolating the #-rhodomycin V from the fermented culture by extracting with at least one solvent at a pH of 6À5 to 7À5. The antibiotic is a red crystalline solid melting with decomposition at 173-175‹ C., soluble in methanol, butanol, acetone, chloroform and aqueous acids, sparingly soluble in water and benzene and insoluble in cyclohexane, and having an optical rotation [α]<SP>20</SP> 313nm = -40 Œ2 degrees (C=0À2 in CHCl 3 ). The fermented culture medium is treated with kieselguhr, filtered and the filter cake is extracted with acetone, the solution evaporated to low volume and back-extracted with butanol, which extract is in turn evaporated and the residue is taken up in chloroform. The culture filtrate also is extracted, preferably with butanol, the extract evaporated and the residue is taken up in chloroform. The antibiotic is obtained by chromatographing the combined chloroform solutions on carboxymethylcellulose with 10% aqueous potassium hydrogen phosphate, making the eluate slightly alkaline (pH 8À0) with ammonia and extracting with chloroform, and precipitating from this extract by adding petroleum ether. It may be purified by chromatography on cellulose with butanol-phosphate buffer 1 : 1, pH 5À8, extracting the alkalized extract with chloroform and precipitating with cyclohexane at boiling temperature. Pharmaceutical compositions having antibiotic activity against Gram-positive and Gramnegative bacteria and mycoplasma organisms (PPLO) comprise the antibiotic #-rhodomycin V of Formula (I) together with a pharmaceutically acceptable carrier, or in unit dosage form optionally without a carrier. The antibiotic may be admixed with an animal feedstuff to provide a veterinary composition.
