Abstract
1,135,522. Liquid chromatography. E. V. PIEL. 30 Aug., 1966 [15 Sept., 1965; 22 Dec., 1965], No. 38557/66. Heading B1X. In liquid column chromatography the chromatography bed consists of particles which have an average size of less than 1.5 microns, preferably less than 1 micron. The liquid mixture to be separated is caused to flow rapidly, preferably by centrifugal force, through the column and is separated by partition between the bed and a moving liquid solvent which is passed through the bed. In operating the centrifuge bucket apparatus of Fig. 1 a bed of particles is established by adding a slurry of particles and a solvent into tube 12 via reservoir 19, the bed being compacted by centrifuging the tube. A sample to be resolved is added to the top surface of the bed and the tube centrifuged thereby establishing chromatographic bands in bed 13 which can be identified visually or 'by elution. With the continuously operating apparatus of Figure 3 the particles e.g. a gel or thixotropic particles are introduced through line 64 and a particle bed is distributed into vessel 50 by distributors 67 and 68 and flows towards the axis of rotation as the vessel is centrifuged. As the particles flow radially inwardly they come into contact with the components of the mixture being distributed through distributors 62 and 63. One or more components flow with the bed for removal at 53 while the other components flow with the developing solvent distributed by distributors 57 and 58 in the opposite direction for removal through exits 51 and 52, materials found to be particularly suitable include silica gels, microfine silica gels, silica aerogels, aluminas, adsorbent carbons, starches, dextrins 2 and finely divided plastics such as nylon.
