Abstract

1,128,303. Centrifugal separators. IVAN SORVALL, Inc. 28 Jan., 1966 [3 Feb., 1965], No. 4022/66. Heading B2P. Biological, e.g. blood, cells are washed by mixing a suspension of the cells with a washing solution having a density less than that of the cells, subjecting the mixture to a centrifuging action thereby providing a counterflow between the cells and the washing solution and removing from the mixture effluent produced during centrifuging. Blood and saline washing solution are continuously drawn by means of a pump 31 respectively from a sterilized plastic bag 21 and a sterilized storage bottle 22, Fig. 1, through a junction fitting 33 where they are mixed and the mixture is delivered via a tube 29, a flow meter 34, tubes 36, 37, 38 and a rotary joint to radially and downwardly extending tubes 44 in a continuous flow centrifuge 39 which is housed in a temperaturecontrolled chamber 47. Each tube 44, Fig. 2, is provided with an inlet tube 48, for the supply of the mixture, and an outlet tube 49, for the removal of effluent, which extend through a compression element 51 closing the tube 44. The tubes 48 and 49 of all tubes 44 are connected respectively to a common inlet and a common outlet manifold (not shown). Each tube 48 is constricted at its outlet end to form one or two terminal ports 57 and is provided along its length with ports 58 which distribute the incoming mixture to spacedapart regions of the tube 44 having different values of centrifugal acoeleration. During centrifugation, the heavier blood cells move outwardly and downwardly in each tube 44, while the lighter washing solution moves in a counterflow direction inwardly and upwardly and emerges as an effluent through outlet tube 49 to be continuously discharged to waste via tubes 41, 42, 43. After completion of a washing operation, each tube 44 is removed from the centrifuge. An air filter 61 is attached by a sterilized tube 62 to the tube 48 and a hollow sterilized needle 63 is connected by a sterilized tube 66 to the tube 49. The tube 44 is then clamped in a support pedestal 67 and the needle 63 is caused to pierce a resilient cap 68 in a storage bottle 59. Transfer of the washed blood cells from the tube 44 to the bottle 59 then takes place by gravity.
