Abstract

1,070,745. Determining the haemoglobin content of blood electrically. H. G. NÍLLER. May 26, 1964 [May 29, 1963], No. 21808/64. Heading G1N. In an apparatus for determining the haemoglobin content of blood, a blood specimen is mixed with a known excess amount of a strong acid (e.g. N/100 hydrochloric acid) and the loss of conductivity due to the reaction of a portion of the acid with the haemoglobin is correlated with the amount of haemoglobin originally present. The apparatus includes a square wave generator 11 (as shown) feeding a bridge circuit via a centre tapped transformer 12. One arm of the bridge is provided by the resistance of the test mixture 2 between a pair of electrodes 4, 5 of a dip cell 3, when it is inserted in the mixture container 1. Another arm of the bridge is formed by a resistor assembly 13 which includes a negative temperature coefficient (N.T.C.) resistor to provide temperature compensation and arranged in thermal contact with the container 1. The unbalance output of the bridge is amplified by amplifier 14 which is coupled to a meter 16 calibrated in grams haemoglobin/100 millilitres of blood. Any electrolyte which will form a practically non-conductive reaction product with haemoglobin may be used such as soluble cyanides e.g. sodium and potassium cyanide, or acids such as hydrogen iodide, hydrogen azide or hydrogen sulphide, as an alternative to hydrochloric acid.
