Abstract

1,051,309. Analysis of peptides. TECHNICON CHROMATOGRAPHY CORPORATION. Sept. 18, 1964 [Oct. 8, 1963], No. 38107/64. Heading BIX. Successive peptides eluted from a chromatography column 10 are first mixed with NaOH solution and heated to hydrolyse them and subsequently quantitatively analysed by a colorimetric technique, in a continuous process. Eluting liquid of progressively varying concentration is delivered from a multi-chamber 'variable gradient device' 12 by a piston pump 14, and the eluted stream is mixed at 52 with NaOH from tube 48 and segmentising bubbles of nitrogen from tube 50, the fluids being delivered by peristaltic pump 46. The mixture is heated in 120 feet of P.T.F.E. tubing 58 in a liquid bath 60 at 95‹C., to facilitate hydrolysis. A capillary tube 62 of P.V.C. continually vents some NaOH to prevent pressure build up. Nitrogen bubbles are vented at 74, and the hydrolysed peptides pass through pump 46', which may be similar to or an extension of pump 46. Acetic acid is added by tube 78 to neutralise excess NaOH; segmentising nitrogen enters the stream from tube 80. A colourdeveloping mixture of methyl cellosolve, ninhydrin and hydrindantin is added at 84 and the stream is heated in bath 88 (at 95‹C.), cooled in coil 90, the nitrogen bubbles are either drawn off or allowed to escape at 103 and the liquid passes through either one colorimeter cell 94, as shown, or through several cells operating at different wavelengths.
