Abstract

1,087,301. Chromatography. TECHNICON CHROMATOGRAPHY CORPORATION. April 1, 1965 [April 8, 1964], No. 13776/65. Heading B1X. [Also in Division B8] In the chromatographic separation of amino acids in an ion exchange column with a downwardly moving developing and eluting buffer solution, the sample is supplied to the top of the column in a liquid having a higher density than said buffer liquid, which flows through said buffer liquid to form a layer on the surface of the resin. The heavier liquid may comprise a 50 g./l. aq. sucrose solution to one volume of which may be added three volumes of sample. An apparatus for applying the heavy solution (see Fig. 1) comprises a housing 12 supportable on column 10 by clamps 18, said housing comprising a first passage 22 for admission of buffer solution, a second passage 38 in the form of a capillory tube or hypodermic needle for admission of the heavy solution into the buffer solution near the surface 43 of resin and a third passage 24 to permit buffer solution to flow out when heavy liquid is injected. The latter two passages may be sealed with closures 34, 42 of special type (see Fig. 2, not shown). The heavy solution is admitted to passage 38 from a syringe.
