Abstract

A process for obtaining BCG cultures comprises cultivating BCG under aerobic conditions in a nutrient solution containing glycerol, a source of nitrogen, inorganic salts and a wetting agent of the "TWEEN" (Registered Trade Mark) type. The aeration may be performed with 0.1 to 0.5 part by volume of air by volume of nutrient solution per minute. The BCG for cultivating in the said nutrient may be prepared by a process wherein a BCG culture is lyophilised, the lyophilisate is stored for a considerable time at the highest storage temperature of the vaccine, and then suspended in an aqueous solution, the solution plated on a solid nutrient medium in such a concentration that separate colonies grow, a few single cell colonies are propagated separately in a liquid medium, the single-cell cultures so obtained are lyophilised and in order to determine the rate of survival a few ampoules of each single-cell culture are plated on a solid medium and the single-cell cultures having the highest rate of survival are used as starting material. The nutrient medium comprises glycerol, "TWEEN" 80 (Registered Trade Mark), L-asparagine, Bacto-Casiton, potasium phosphate sodium phosphate, ferric ammonium citrate, magnesium sulphate, calcium chloride, zinc sulphate, copper sulphate and deionised water and albumin solution or sodium acetate and "Difco" (Registered Trade Mark) yeast extract.
