Abstract

The invention comprises 4-R4-3-R3-1-R1-isoquinolines wherein R1 is halo or optionally substituted amino, R3 is optionally substituted amino-methyl, and R4 is lower (1-5 C) alkyl, and the carbocyclic portion of the isoquinoline nucleus may be substituted by halogen, lower alkoxy, lower alkyl, trifluoromethyl, nitro and/or amino; their salts; and their preparation by (1) reaction of 4-R4-3-halomethyl - 1 - haloisoquinolines with an appropriate amine or two amines in succession (diisoquinolylmethyl - amines may result); (2) the interconversion of products by nitration, reduction of nitro to amino, or replacement of amino by halo. Isoquinoline derivative starting materials prepared are: 1-chloro-3-chloromethyl-4-methyl-5 - nitroisoquinoline, 4,4 - dimethyl - 7 - nitro, - amino, and - chloro - homophthalimide, and 1,7 - dichloro - 3 - chloromethyl - 4 - methylisoquinoline. Pharmaceutical compositions incorporate compounds of the invention, forms for oral and parenteral administration as antitussives being described.
