Description
The present invention relates to the field of valorization of moabi cake. More specifically, the invention relates to an extract of hydrolyzed moabi cake as well as its use in cosmetics, in particular as an agent for preventing and/or treating the signs of skin aging, for hydrating the skin and for whitening, lightening and/or depigmenting. the skin.
The skin is the human body's main protective barrier against external aggressions such as atmospheric pollution, climatic variations and UV radiation.
The skin is made up of three layers: the epidermis, the dermis and the hypodermis. The epidermis, which is in contact with the outside, is particularly affected by external aggressions. The epidermis is covered by a hydrolipidic film and is composed of four layers: the stratum corneum, the granular layer, the spinous layer and the basal layer. The skin, and in particular the epidermis, is constantly subjected to stress which causes it to age.
We generally distinguish between intrinsic aging and extrinsic aging.
Intrinsic aging, also called physiological aging, causes a slowdown in the renewal of keratinocytes which are the cells of the superficial layers of the skin, particularly the epidermis. The reduction of subcutaneous adipose tissue and the appearance of fine wrinkles are the main manifestations of intrinsic aging.
Extrinsic aging is characterized by a loss of skin elasticity, an alteration of the skin's barrier function and the appearance of deep wrinkles. Extrinsic aging is essentially due to repeated exposure of the skin to the sun; it is also associated with the appearance of darker spots which generate areas with a non-homogeneous appearance. This aging is also called photoaging or actinic aging.
The present invention aims more particularly at intrinsic aging.
These dark spots are usually due to a localized concentration of melanin on the surface of the skin. Melanin is the pigment responsible for skin color; it is produced by melanocytes located in the epidermis.
Tyrosine → DOPA → Dopaquinone → L eucodopachrome → D opachrome → M élanine The mechanism of formation of skin pigmentation, that is to say melanin, is particularly complex and schematically involves the following main stages, at the level of the melanocytes of the basal layer:
Tyrosine → DOPA → Dopaquinone → L eucodopachrome → D opachrome → M lanin
Tyrosinase (monophenol dihydroxyl phenylalanine: oxygen oxidoreductase EC 1.14.18.1) is the essential enzyme involved in this series of reactions. It catalyzes in particular the reaction of transformation of tyrosine into Dopa (dihydroxyphenylalanine) thanks to its hydroxylase activity and the reaction of transformation of Dopa into dopaquinone thanks to its oxidase activity.
There therefore still remains a need for new cosmetic compositions intended to be applied to the skin and which have moisturizing, anti-aging and depigmenting properties.
The inventors showed that a solution to this technical problem could be provided using a particular extract of moabi cake.
Moabi or Baillonella toxisperma is a plant species in the Sapotaceae family. The moabi is a large tree growing in the tropical rainforests of Africa. It is traditionally exploited as tropical wood. The seeds of moabi fruits are traditionally pressed to obtain an oil used in the local cosmetic industry.
Moabi oil, obtained by pressing and then more or less extensive refining, generates a large quantity of solid by-products called moabi cakes.
There still remains a need to find new ways of valorizing moabi meal with high added value.
Surprisingly and advantageously, the inventors have demonstrated a particular extract of moabi cake obtained by a particularly innovative alkaline hydrolysis process in the treatment of moabi cake, and presenting advantageous effects in cosmetics when applied to the skin, more particularly cosmetic properties, notably moisturizing, anti-aging and depigmenting.
The subject of the present invention is a moabi meal hydrolyzate comprising 0.50 to 1.50% by weight, preferably 0.83% by weight, of proteins relative to the total weight of the hydrolyzate; from 0.50 to 2.00% by weight, preferably 1.19% by weight, of sugars relative to the total weight of the hydrolyzate and from 15 to 30 ppm, preferably 23 ppm, of polyphenols relative to the total weight of the hydrolyzate.
Avantageusement ledit hydrolysat de tourteau de moabi est tel que son spectre UV visible présente un pic à une longueur d’onde λ de 230 nm pour une absorbance de 2,262.Advantageously, said moabi cake hydrolyzate is such that its visible UV spectrum has a peak at a wavelength λ of 230 nm for an absorbance of 2.262.

Thus the hydrolyzate of moabi cake and the cosmetic composition according to the invention advantageously make it possible to prevent and/or reduce the signs of skin aging, in particular the loss of elasticity of the skin, the alteration of the barrier function of the skin, the appearance of wrinkles and/or fine lines. It has been observed that treated skin is firmer.
The hydrolyzate of moabi cake and the cosmetic composition according to the invention also make it possible to whiten and/or lighten and/or depigment the skin of the face and/or body in a lasting manner. They also help reduce the size of pigmented brown spots and therefore make specific areas of the face and/or body more homogeneous.

HydrolysatHydrolyzate
The hydrolyzate according to the present invention is prepared from moabi or Baillonella toxisperma seed cakes.
It is known that moabi seeds are used for the extraction of their oil, useful for self-consumption or in various traditional medicinal treatments. The cake obtained after delipidating (de-oiling) the seed is a waste that is currently little recovered.
According to a preferred embodiment, the hydrolyzate according to the invention is obtained from the cake of shelled moabi seeds.
According to the present invention, the moabi cakes are recovered after extracting the oil from the seeds and then hydrolyzed.
The hydrolyzate according to the invention comprises from 0.50 to 1.50% by weight, preferably 0.83% by weight of proteins relative to the total weight of the hydrolyzate, from 0.50 to 2.00% by weight. preferably 1.19% by weight of sugars relative to the total weight of the hydrolyzate and from 15 to 30 ppm, preferably 23 ppm of polyphenols relative to the total weight of the hydrolyzate.
- fournir des graines de moabi ;
- délipider les graines de moabi et récupérer le tourteau ;
- placer le tourteau de moabi en phase aqueuse ;
-ajouter un acide faible de manière à ajuster le pH à une valeur allant de 4,5 à 5,5 ;
- effectuer une séparation solide/ liquide,
- récupérer l’hydrolysat obtenu.As already mentioned, the process for preparing moabi cake hydrolyzate comprises at least the following steps, in this order:
- provide moabi seeds;
- remove the fat from the moabi seeds and collect the cake;
- place the moabi cake in the aqueous phase;
- subject the cake to at least one hydrolysis step using an alkaline solution at a pH ranging from 13 to 14 for a period ranging from 30 minutes to 4 hours,
-add a weak acid so as to adjust the pH to a value ranging from 4.5 to 5.5;
- carry out a solid/liquid separation,
- recover the hydrolyzate obtained.
Generally, the seeds intended to be used in the process according to the invention are shelled, that is to say the almond is used.
The moabi cake used in the process according to the invention is obtained after delipidation of the moabi seeds, preferably moabi kernels. This step aims to extract the oil from the moabi seeds.
Delipidation can be carried out by mechanical pressing of the seeds or by extraction using a non-polar organic solvent, for example hexane.
De préférence les graines éventuellement sous forme d’amandes sont séchées ou laissées sécher de manière à obtenir un taux d’humidité inférieur à 7%.Preferably the seeds, possibly in the form of almonds, are dried or left to dry so as to obtain a humidity level of less than 7%.
Then they are subjected to delipidation so as to separate the oil from the rest of the seed and to obtain the cake. Generally the cake contains less than 5% by weight of oil, called “residual oil”, relative to the total weight of the cake.
According to a preferred variant, the cake is then crushed mechanically with any type of mechanical crushers such as hammer, flail, knife, crushing/shredding, ball or ball or pestle systems, but also with any type of cryomill. .

The quantity of total proteins was determined by colorimetric assay according to the Lowry method. For this colorimetric assay, a standard curve of the optical density, called OD in the rest of the text, measured at 750 nm, as a function of the quantity of proteins was produced. The reference protein used in this experiment is bovine serum albumin (BSA).

B.Analyse de la teneur des polyphénols totaux de l’extrait par la méthode utilisant le réactif de Folin Ciocalteu b.Analysis of the content of total polyphenols in the extract by the method using the reagent of Folin Ciocalteu
This analysis was carried out using the extract from Example 1 diluted 1/10 in distilled water.
The determination of total polyphenols was carried out according to the method described in 1965 by Singleton and Rossi using the Folin-Ciocalteu reagent. Folin Ciocalteu's reagent is a yellow acid made up of a mixture of phosphotungstic acid (H3PW12O40) and phosphomolybdic acid (H 3 PMo 12 O 40 ). It is reduced, during the oxidation of phenols, to a mixture of blue oxides of tungsten and molybdenum (Ribereau, 1968). The coloring produced, including the maximum absorption at 720nm, is proportional to the quantity of polyphenols present in the sample.
For this colorimetric assay, a standard curve of the OD, measured at 720 nm, as a function of the quantity of polyphenols was produced.
The reference compound used in this experiment is gallic acid.

Après incubation pendant 30 minutes à 45°C, les mesures de DO à 720 nm ont été réalisées.6 standard solutions are prepared by taking 400 µL from each of tubes 1 to 6 in Table 3 to which 2000 µL of Folin Ciocalteu reagent and 1600 µL of 7.5% Na 2 CO 3 solution are added. After incubation for 30 minutes at 45°C, OD measurements at 720 nm were carried out.
Les mesures de DO à 720 nm ont permis de définir une courbe d’équation :y = 0,0967x - 0,019 avec R² = 0, 9982. The DO measurements at 720 nm made it possible to define an equation curve: y = 0.0967x - 0.019 with R² = 0.9982.
Then the determination of the quantity of total polyphenols in each hydrolyzate sample to be tested (sample tested in triplicate) was obtained by preparing samples to be tested according to the same protocol as that used for the standard range and then reporting the OD values. obtained for the samples to be tested on the standard curve.

Analysis of the sugar content of the hydrolyzed extract by the Dubois method
The quantity of total sugars (saccharides) was determined by colorimetric assay according to the Dubois method. This method is based on the formation of a chromogenic product of sugar degradation by the action of a strong acid into furfural derivatives. These chromogenic products are then condensed with phenol to obtain an orange-colored chromophore.
For this colorimetric assay, a standard curve of the OD, measured at 490 nm, as a function of the total quantity of sugars was produced. The reference product used in this method is glucose.
Then the determination of the total quantity of sugars in each hydrolyzate sample to be tested was obtained by preparing samples to be tested according to the same protocol as that used for the glucose standard range and then reporting the OD values obtained for the samples. to be tested on the standard curve.
